Thames Valley Police and South Central Ambulance Service were called to a woodland area between Childs Way and Padstow Avenue on the Fishermead estate at around 3.20pm on Wednesday.
There had been reports of an altercation between a group of men, during which a man had been stabbed, police said.
The victim, a man aged in his 20s, died at the scene.
His next of kin have been informed.
Officers said five men have been arrested on suspicion of murder in connection with the incident and remain in police custody.
Detective Inspector Mike Roddy, of Thames Valley Police major crime unit, said a police cordon was in place and asked the public to avoid the area.
He said: "Thames Valley Police is in the early stages of this murder investigation after a man has tragically died.
"Our priorities at this time are to conduct a thorough investigation into this incident and support those who have been affected.
"Officers are currently at the scene, which has been cordoned off and we would ask members of the public to avoid the area."
He asked anyone who witnessed any suspicious activity in the area or with any mobile phone, dash-cam or CCTV footage to contact police.
"I understand that this incident will have caused concern within the community, but please be assured that the major crime unit has commenced an extensive investigation," he added.
